Tesofensine – Research Grade Triple Reuptake Inhibitor

Tesofensine is a novel triple monoamine reuptake inhibitor that blocks the reabsorption of serotonin, norepinephrine and dopamine in the brain. By increasing the availability of these neurotransmitters, tesofensine acts on central appetite and energy regulation pathways instead of gut hormones. Originally investigated for Parkinson’s and Alzheimer’s disease, it was repurposed for obesity when clinical trials consistently showed significant weight reduction.

Phase 2 trials using a once‑daily 0.5 mg (500 mcg) oral dose produced an average 11.3 kg weight loss (~10.6 % of body weight) over 24 weeks. This dual action—reducing cravings while potentially increasing resting metabolic rate—distinguishes tesofensine from GLP‑1 agonists.

Tesofensine remains investigational; phase 3 clinical trials are ongoing and it is not FDA‑approved. Reported side effects include dry mouth, headache, insomnia, nausea and constipation; it may also increase heart rate.
